Other provisions of the law still go into effect on Thursday, such as the portion of the law that creates misdemeanors crimes for harboring and transporting illegal immigrants.
Arizona Governor Jane Brewer said she would appeal the ruling.
Unions and religious groups in Los Angeles say the fight against the Arizona law is not over and they want to show their willingness to continue the fight.
"People that come here deserve rights. They deserve equality. They don't deserve to be harassed," protester David Feldman told reporters before heading toward Arizona.
Brewer signed the law, Senate Bill 1070, in April. The law makes it a crime to stay in the country illegally and empowers local law enforcement in Arizona to check the immigration status of people suspected of staying in the country illegally. But the measure specifically bars police from racial profiling.
